DEVOTION Camptothecin is a plant secondary metabolite used as an anti-cancer drug that damages DNA, leading to the destruction of the cell. It comes from Camptotheca acuminata, a deciduous tree found in southern China. A close analogue, 9-methoxycamptothecin, is also present in the same source.

Mechanism of Action:

Camptothecin affects the activity of the enzyme topoisomerase I, whose normal action is to cleave, unwind, and religate DNA. When camptothecin binds to topoisomerase I, it will be able to cleave but not to religate DNA. Thereby, camptothecin causes single strand breaks in DNA. 

Function:

-Treatment Gastrointestinal cancer and leukemia,

-Plant growth regulator activity and kill insects. 

-Treatment of gastric cancer, liver cancer, head and neck cancer and leukemia,, with fewer side effects than camptothecine.

Application:

1. Camptothecin and its derivatives are unique in their ability to inhibit DNA Topoisomerase I, by stabilizing a covalent reaction intermediate termed the cleavable complex which ultimately causes tumor cell death. 

2. In clinical it is widely believed that camptothecin analogs exhibited remarkable anti-tumour and anti-leukaemia activity. Topoisomerase is a basilic enzyme in the process of DNA replication, it is responsible for the winding / unwinding of the supercoiled DNA composing the chromosomes. If the chromosomes cannot be unwound, transcription of DNA message cannot occur and the protein cannot be synthesized, it ultimately causes cell death. 

3. Application of camptothecin in clinic is limited due to serious side effects and poor water-solubility. 

4. At present, some camptothecin analogs,either semi-synthetic or synthetic drug based on camptothecin, have been applied cancerous therapy such as topotecan and irinotecan while others have been obtained satisfying curative effects in clinic.and insomnia. Do not take horny goat weed if you are taking heart medicines.
